Structure of the autoinhibited Parkin catalytic domain

Parkin is kept in an autoinhibited conformation by two key interactions. First, the linker connecting the IBR and the RING2 blocks the conserved E2-Ub binding sight of RING1. Second, RING2 forms a hydrophobic interface with RING0 burying C431. This structure further elucidates the possible mutations which could cause Parkinson's disease.
