B-catenin interactions with Tcf
B-catenin is also a key signaling molecule in the canonical Wnt pathway that plays diverse and critical roles in embryonic development and in adults. It was recently discovered that is also a critical regulator of stem cells.
Elevated levels of b-catenin can lead to its constitutional activation by the b-catenin-Tcf complex.

There are two sites of interactions between Tcf and b-catenin (R3-R10). Acidic residues (red) at either end of Tcf form salt bridges with basic residues in b-catenin (black).

Tcf4 can specifically recognize beta-catenin using alternative conformations.
