SpCas9 with CONSURF overlay

The color of each residue is determined by its level of evolutionary conservation, based on CONSURF calculations using a similar SpCas9 structure and its 138 closest relatives. Purple represents residues that are highly conserved, white represents residues that are moderately conserved, blue represents residues that are highly variable, and yellow represents residues whose conservation scores could not be calculated. Overall, conservation tends to be greatest in residues around the main channel, and in the catalytic domains.
